چکیده
The present study was designed to examine the hepatoprotective effects of curcumin (CMN), resveratrol (RSV) and ursodeoxycholic acid (UDCA) against ethinylestradiol and chlorpromazine -induced intrahepatic cholestasis. Ethinylestradiol (EE) and chlorpromazine HCl (CPZ) were administered orally once daily simultaneously with the tested compounds for one week. At the end of the experimentation period, animals were sacrificed. Blood and liver samples were obtained, serum was used for the determination of alanine aminotransferase (ALT), aspartate aminotransferase (AST), direct& indirect bilirubin, gamma-glutamyltransferase (γ-GT) and cholesterol. Liver samples were used for hiatopathological examination. The results demonstrate that CMN showed marked reduction in all biochemical indices of cholestasis while RSV failed to ameliorate EE-CPZ-induced changes. UDCA produced limited protective effects against EE-CPZinduced cholestatic injury. This study suggests that CMN has a potent anticholestatic effect in case of EE-CPZ-administration.

Effects of Three Different Fibrates on Intrahepatic Cholestasis Experimentally Induced in Rats
Background. Activation of PPAR α modulates cholesterol metabolism and suppresses bile acid synthesis. This study aims to evaluate the effect of PPAR α agonists, fenofibrate, bezafibrate, and gemfibrozil, on acute cholestasis induced by ethinylestradiol (EE) plus chlorpromazine (CPZ) in rats. Method. 100 male albino rats (150-200 gm) were divided randomly into 10 equal groups. Control group rece...

AIM To explore the effects of matrine (MT) on acute intrahepatic cholestasis induced by 17alpha-ethinyl estradiol (EE) in rats. METHODS Acute intrahepatic cholestasis in rats were induced by EE, and the effects of MT on acute intrahepatic cholestasis were explored and compared with ursodeoxycholic acid (UDCA) by serum biochemical determination and bile excretion experiments. RESULTS The ser...
متن کاملThe Modulatory Effects of Pentoxifylline in Biochemical Changes Induced By 17α-Ethinyl Estradiol in the Rat Model
Background: Ethinylestradiol (EE) has induced cholestasis and hepatotoxicity in animal studies through reducing bile acid uptake by hepatocytes and induce of oxidative stress. Pentoxifylline (PTX) is a drug that by inhibition of release or transcription of proinflammatory cytokine cause prevents oxidative stress of liver cell and reduction of damage. We aimed to evaluate the effects of pentoxif...
متن کاملThe effect of curcumin on altered expression of BAX and BCL2 genes following cholestasis in the frontal region of male rats
Background: Cholestasis occurs as a result of malfunction or secretion of bile; and complications of this disease include apoptosis (planned death). Curcumin is an effective extract of turmeric rhizome that has anti-apoptosis properties. Thus in this study, we investigated the expression of BAX and BCL2 genes following cholestasis and curcumin in the frontal region of male rats. Materials and m...

Background and purpose: Resveratrol (RSV) is a naturally polyphenolic compound found in grapes and other plant sources. It has potent free radical scavenger and antioxidative properties. The aim of this study was to evaluate the protective effect of resveratrol against Perfluorooctanoic acid (PFOA) induced mitochondrial toxicity in isolated liver mitochondria. Materials and methods: Mitochondr...
